Human Chromogranin A ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of human Chromogranin A in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated microplate platform with colorimetric detection read at 450 nm on a microplate reader. A capture antibody specific for Chromogranin A is pre-immobilised on the plate; standards and samples are added and bound analyte is detected using a Biotinylated Anti-Chromogranin A Antibody followed by an HRP-Avidin conjugate. TMB substrate is added and the enzymatic reaction is stopped with acidic stop solution, producing a yellow colour whose absorbance is proportional to Chromogranin A concentration. The assay shows a sensitivity of 23.5 pg/ml, a working range of 62.5-4000 pg/ml, and a total run time of 4 hours 30 minutes. Components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.

Recovery has been validated across serum and plasma matrices. In neat samples, serum (n=5) gave 80-102% recovery with a 91% average, EDTA plasma (n=5) gave 81-100% with a 90% average, and heparin plasma (n=5) gave 80-89% with an 84% average. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types. The assay is a quantitative sandwich ELISA with reactivity limited to human samples.
