From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is designed for the in vitro quantitative determination of human CHRNB3 in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a 96-well microtiter plate pre-coated with an antibody specific for CHRNB3. Standards and test samples are added to the wells, the plate is washed, and a Biotinylated Anti-CHRNB3 Antibody is then added, followed by an HRP-Avidin conjugate. TMB substrate solution visualises the enzymatic reaction, producing a blue-coloured product that turns yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of CHRNB3 captured per well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The quantitative assay has a range of 0.312-20 ng/ml and a sensitivity of 0.125 ng/ml, with a total assay time of 4h 30m on a pre-coated microplate platform. Detection is colourmetric. Recovery was evaluated in serum (n=5; range 83%-99%; average 91%), EDTA plasma (n=5; range 80%-100%; average 90%), and heparin plasma (n=5; range 82%-99%; average 90%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions across the same sample types. Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage at +4°C or -20°C depending on the component.
