Human Cholinergic Receptor, Nicotinic, Beta 2 ELISA Kit

A sandwich enzyme immunoassay from Antibodies.com enables the in vitro quantitative determination of human Nicotinic Acetylcholine Receptor beta 2 (CHRNB2) in tissue homogenates or other biological fluids. An antibody specific for the target is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, followed by a biotinylated anti-CHRNB2 detection antibody, an HRP-avidin conjugate and TMB substrate. The HRP-catalysed reaction yields a blue product that turns yellow after addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader.

The assay has a stated product range of 0.156-10 ng/ml and a sensitivity of 0.054 ng/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.

Spike-and-recovery testing (n=5 per matrix) gave average recoveries of 91% in serum (range 83% - 99%), 90% in EDTA plasma (range 80% - 100%) and 90% in heparin plasma (range 82% - 99%). Linearity assessed at dilutions of 1:2, 1:4, 1:8 and 1:16 returned 87-109%, 74-104%, 90-94% and 86-108% in serum; 86-101%, 90-101%, 78-110% and 84-96% in EDTA plasma; and 90-101%, 77-107%, 93-96% and 89-109% in heparin plasma.
