Human Cholecystokinin B Receptor ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Cholecystokinin B Receptor (CCK2-R) in tissue homogenates or other biological fluids. The assay uses a pre-coated microplate on which an antibody specific for CCK2-R is immobilised. Standards and test samples are added to the wells, where CCK2-R is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-CCK2-R Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then added to visualise the HRP enzymatic reaction; the resulting blue product turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of CCK2-R captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The kit has a sensitivity of less than 0.052 ng/ml and an assay range of 0.156–10 ng/ml. Total assay time is 4h 30m, and detection is colourimetric. Components supplied include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Storage instructions indicate that kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Validation data are provided for human serum, EDTA plasma, and heparin plasma (n=5 each). Reported average recoveries are 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%), and 84% for heparin plasma (range 80%–89%). Linearity upon dilution has been evaluated at 1:2, 1:4, 1:8 and 1:16 sample dilutions.
