Description

This sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human CLIC1 in tissue homogenates, cell lysates, and other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for CLIC1, into which standards and test samples are added. CLIC1 present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-CLIC1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualize the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of CLIC1 captured per well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The kit demonstrates a sensitivity of <0.123 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Recovery has been validated across sample types: serum (range 80–102%, average 91%), EDTA plasma (range 81–100%, average 90%), and heparin plasma (range 80–89%, average 84%). Dilution linearity was assessed in the same sample matrices at 1:2, 1:4, 1:8, and 1:16 dilutions. The platform is a pre-coated microplate arranged as 12 x 8 well strips.

Kit components include the pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
