Human Chitinase 3 Like Protein 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Chitinase 3 Like Protein 2 (YKL-39) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microplate in which a specific capture antibody immobilises the target, followed by incubation with a biotinylated anti-YKL-39 detection antibody and an HRP-avidin conjugate. A TMB substrate is then added and the enzymatic reaction is stopped with an acidic solution; the resulting colour intensity, read at 450 nm, is proportional to the YKL-39 concentration and is interpolated from a standard curve.

The kit reports a sensitivity of less than 1.33 ng/ml and a working range of 3.12–200 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Recovery has been characterised in serum, EDTA plasma and heparin plasma. Sample collection guidance covers serum (clotted for 30 minutes at room temperature and centrifuged at 1000 × g for 15 minutes), plasma (EDTA or heparin as anticoagulant, centrifuged at 1000 × g for 15 minutes within 30 minutes of collection), tissue homogenates (1 g tissue per 9 ml PBS, centrifuged at 5000 × g for 5 minutes) and cell lysates; the total protein concentration of each well sample should not exceed 0.3 mg.

Kit components include a pre-coated microplate, two vials of lyophilised standard, Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and adhesive plate sealers.
