Human Chemokine C-X-C-Motif Receptor 4 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human CXCR4 (Chemokine C-X-C-Motif Receptor 4) in tissue homogenates, cell lysates, and other biological fluids.

The 96-well microplate is pre-coated with an antibody specific for CXCR4. Standards and samples are added to the wells, where CXCR4 binds to the immobilised capture antibody; a biotinylated anti-CXCR4 detection antibody is then added, followed by an HRP-avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of CXCR4 captured, and concentration is calculated from the absorbance read at 450 nm against a standard curve. The reported assay range is 0.156–10 ng/ml, with a sensitivity of <0.060 ng/ml, and the total assay time is approximately 4 hours 30 minutes.

Reported sample recovery averages 91% for serum (range 80%–102%), 90% for EDTA plasma (81%–100%), and 84% for heparin plasma (80%–89%). Linearity has been evaluated across 1:2 to 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the kit is suitable for use with cell lysates and tissue homogenates.
