The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for CCT2 pre-coated onto a 96-well microplate (12 x 8 well strips). Standards and test samples are added to the wells; CCT2 is captured by the immobilized antibody, then detected with Biotinylated Anti-CCT2 Antibody and HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow after addition of the acidic stop solution, and absorbance is read at 450 nm in a microplate reader against a standard curve.

Sensitivity is <0.067 ng/ml, with an assay range of 0.156-10 ng/ml. Typical recovery is 80-102% (average 91%) in serum, 81-100% (average 90%) in EDTA plasma, and 80-89% (average 84%) in heparin plasma, across dilution series of 1:2, 1:4, 1:8, and 1:16. Total assay time is 4 hours 30 minutes.
