A sandwich ELISA developed by Antibodies.com for the in vitro quantitative determination of human Cerberus (Cer1) in serum, plasma, tissue homogenates and other biological fluids. The format uses a pre-coated 96-well microplate (12 x 8 well strips) with an antibody specific for Cer1 immobilised on the wells. Standards and samples are added, followed by a Biotinylated Anti-Cer1 Antibody and an HRP-Avidin conjugate. A TMB substrate is then added and the enzymatic reaction is stopped with an acidic stop solution, producing a yellow colour read at 450 nm in a microplate reader. The colour intensity is proportional to the amount of Cer1 captured in each well, and concentration is calculated against the standard curve.

Quantitative sandwich ELISA with colourmetric detection. Reported sensitivity is <12.3 pg/ml and the working assay range is 31.2-2000 pg/ml. Total assay time is 4h 30m. Spiked recovery in serum (n=5) averages 91% with a range of 80-102%, in EDTA plasma averages 90% (81-100%), and in heparin plasma averages 84% (80-89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.

Supplied components include the pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4°C or -20°C as indicated in the supplied instructions, and the kit should not be used past its stated expiration date.
