Human Cathepsin B ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human Cathepsin B in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Cathepsin B pre-coated onto a microplate. Standards and samples are added to the wells, where Cathepsin B present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-Cathepsin B Antibody binds the captured analyte, followed by addition of an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and Cathepsin B concentration is calculated against a standard curve.

Reported sensitivity is <0.119 ng/ml with an assay range of 0.312–20 ng/ml. The total assay time is 4 hours 30 minutes. Recovery performance has been documented in serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5) and heparin plasma (range 80–89%, average 84%, n=5). Linear dilution performance is reported across 1:2 to 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
