Human Caspase 9 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Caspase-9 in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Caspase-9 is also referred to by the synonyms APAF-3, Apoptotic protease Mch-6, CASP-9, CASP9, ICE-LAP6 and MCH6, and is associated with the descriptive names apoptotic protease-activating factor 3 and ICE-like apoptotic protease 6.

The assay uses a pre-coated microplate bearing an immobilised antibody specific for Caspase-9, allowing standards and samples to bind to the wells. Detection is performed using a biotinylated anti-Caspase-9 antibody followed by an HRP-avidin conjugate. The enzymatic reaction is visualised with a TMB substrate that changes from blue to yellow after addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of Caspase-9 is calculated by reference to the standard curve. The total assay time is 4 hours 30 minutes.

According to the manufacturer, the product range is 0.156–10 ng/ml and the sensitivity is less than 0.052 ng/ml. Reported spike-and-recovery values are: serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%) and heparin plasma 80%–89% (average 84%). Components are stored at +4°C or −20°C as instructed; the kit is shipped at +4°C and should not be used past its stated expiration date.
