Human Caspase 6 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of human Caspase-6/CASP-6 in serum, plasma, tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate in a 12 x 8 well strip format with a colorimetric detection method read on a standard microplate reader.

In the procedure, an antibody specific for Caspase-6/CASP-6 is immobilised on the plate. Standards and test samples are added, and the target in each sample is captured by the coated antibody. Following incubation and washing, a biotinylated anti-Caspase-6/CASP-6 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction; after addition of an acidic stop solution, the resulting yellow colour is read at 450 nm, and the concentration of Caspase-6/CASP-6 is calculated from a standard curve.

The assay has a reported product range of 0.156–10 ng/ml and a sensitivity of <0.069 ng/ml, with a total assay time of 4 h 30 min. Recovery in serum (n=5) ranged from 80–102% with an average of 91%; EDTA plasma (n=5) ranged from 81–100% with an average of 90%; and heparin plasma (n=5) ranged from 80–89% with an average of 84%. Linearity has been demonstrated at dilutions of 1:2, 1:4, 1:8, and 1:16 in serum, EDTA plasma, and heparin plasma. Kit components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
