Human Casein Alpha ELISA Kit

A sandwich ELISA kit from Antibodies.com designed for the in vitro quantitative determination of human Casein in tissue homogenates, cell lysates, cell culture supernates, breast milk and other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate. Standards and samples are added to wells coated with an antibody specific for Casein; after washing, a Biotinylated Anti-Casein Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow on addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader.

The kit offers a product range of 1.56–100 ng/ml and a sensitivity of <0.55 ng/ml, with a total assay time of 4 h 30 m. Sample recovery has been evaluated across 1:2, 1:4, 1:8 and 1:16 dilutions in serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5), with reported ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma at the respective dilutions. Kit components include a pre-coated 96-well microplate (12 × 8 well strips), lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
