Human Cartilage Associated Protein ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human CRTAP/CASP (Cartilage Associated Protein) in tissue homogenates, cell lysates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, in which an antibody specific for CRTAP/CASP is pre-coated onto a microtiter plate. Standards and test samples are added, followed by a Biotinylated Anti-CRTAP/CASP Antibody and an HRP-Avidin conjugate. TMB substrate is then added and catalysis produces a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of CRTAP/CASP is calculated by reference to the standard curve. The reported assay range is 0.156–10 ng/ml and sensitivity is <0.061 ng/ml, with a stated total assay time of 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.

Reported recovery in spiked samples was serum 80–102% (n=5, average 91%), EDTA plasma 81–100% (n=5, average 90%), and heparin plasma 80–89% (n=5, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was reported as serum 87–91% / 87–107% / 74–101% / 92–97% (n=5), EDTA plasma 90–105% / 84–101% / 90–101% / 79–108% (n=5), and heparin plasma 84–95% / 92–105% / 82–105% / 89–91% (n=5).
