Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Carnosine Dipeptidase 1 (CNDP1/CN1) in serum, plasma, cerebrospinal fluid or other biological fluids. The assay uses an antibody specific for CNDP1/CN1 pre-coated onto a 96-well microplate. Standards and samples are added, the wells are washed, and a Biotinylated Anti-CNDP1/CN1 Antibody is added, followed by HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of CNDP1/CN1 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Sensitivity is reported as <1.45 ng/ml with a product range of 3.12–200 ng/ml, and total assay time is 4h 30m. Detection is colourimetric on a pre-coated microplate platform (12 x 8 well strips). Sample recovery has been demonstrated in serum (n=5) at 80–102% (average 91%), in EDTA plasma (n=5) at 81–100% (average 90%), and in heparin plasma (n=5) at 80–89% (average 84%). Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

The kit is suitable for serum and plasma, with dilution linearity data reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. An easy-to-follow protocol is provided.
