Human Cardiotrophin Like Cytokine Factor 1 ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human NNT1 (Cardiotrophin Like Cytokine Factor 1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a biotinylated anti-NNT1 detection antibody and an HRP-avidin conjugate for signal generation.

NNT1 present in standards or test samples is captured by the immobilised antibody on the plate wells. After incubation and washing, the Biotinylated Anti-NNT1 Antibody binds the captured analyte, followed by addition of HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour, read as O.D. absorbance at 450 nm in a colorimetric microplate reader, is proportional to the amount of NNT1 captured, and the concentration is calculated against a standard curve.

Reported performance characteristics include a sensitivity of 13.9 pg/ml and a quantitative range of 31.2-2000 pg/ml, with a total assay time of 4h 30m. Spike-and-recovery values were measured in serum (n=5, range 80%-102%, average 91%), EDTA plasma (81%-100%, average 90%) and heparin plasma (80%-89%, average 84%). Linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across the same three sample types.
