This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human CD66b in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for CD66b that is pre-coated onto a 96-well microplate in a 12 x 8 well strip format. Standards and test samples are added to the wells; CD66b present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-CD66b Antibody binds the captured CD66b, followed by addition of an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of CD66b captured in each well, read as O.D. absorbance at 450 nm in a microplate reader.

The assay shows a sensitivity of <5.9 pg/ml and an assay range of 15.6–1000 pg/ml, with a reported assay time of 4h 30m. Sample recovery figures from the manufacturer include serum 80–102% (average 91%), EDTA plasma 81–101% (average 91%), and heparin plasma 80–89% (average 84%). Components include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, stored at +4°C or −20°C as instructed. The kit is read using a colorimetric microplate reader and is intended for research use only.
