A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Carcinoembryonic Antigen (CEACAM5) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit employs the sandwich enzyme immunoassay technique using an antibody specific for CEACAM5 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-CEACAM5 Antibody and HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The concentration of CEACAM5 is calculated from the O.D. absorbance at 450 nm against a standard curve.

The reported product range is 78.1-5000 pg/ml with a sensitivity of <31.2 pg/ml, read on a colorimetric microplate reader, with a total assay time of 4h 30m. Kit components include a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers, to be stored at +4°C or -20°C as instructed and not used past the expiration date.

Reported recovery ranges are 80-102% for serum (average 91%), 81-100% for EDTA plasma (average 90%) and 80-89% for heparin plasma (average 84%), with dilution linearity data provided across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma.
