Human Carboxypeptidase M ELISA Kit

Offered by Antibodies.com, this assay is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of human Carboxypeptidase M (CPM) in serum, plasma, tissue homogenates, or other biological fluids. It uses a 96-well microplate pre-coated with an antibody specific for CPM. Standards and samples are added to the wells so that CPM is captured by the immobilised antibody; after washing, a biotinylated anti-CPM antibody is added, followed by an HRP-avidin conjugate. TMB substrate then visualises the HRP reaction as a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and CPM concentration is calculated by reference to the standard curve.

The stated assay range is 78.1-5000 pg/ml with a sensitivity of 28 pg/ml (also reported as <28 pg/ml), and the total assay time is 4 hours 30 minutes. Reported spike-and-recovery values are: serum n=5, range 80%-102%, average 91%; EDTA plasma n=5, range 81%-100%, average 90%; heparin plasma n=5, range 80%-89%, average 84%. Dilutional linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 across serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%) and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%), each at n=5.
