Human Carbonic Anhydrase IX ELISA Kit

A sandwich ELISA supplied by Antibodies.com for the in vitro quantitative determination of human Carbonic Anhydrase 9/CA9 in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate (12 x 8 well strips). An antibody specific for Carbonic Anhydrase 9/CA9 is immobilised on the plate; standards and test samples are added and any CA9 present is captured. Following incubation and washing, a Biotinylated Anti-Carbonic Anhydrase 9/CA9 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of CA9 is calculated by reference to the standard curve.

The assay has a stated sensitivity of <6.5 pg/ml and a product range of 15.6–1000 pg/ml. The total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date. Detection is colorimetric.

Recovery data supplied in the product literature for spiked samples (n=5) are: serum range 80%–102% (average 91%), EDTA plasma range 81%–100% (average 90%), and heparin plasma range 80%–89% (average 84%). Dilution linearity is also reported for serum, EDTA plasma and heparin plasma across serial dilutions of 1:2, 1:4, 1:8 and 1:16.
