Human Carbonic Anhydrase III, Muscle Specific ELISA Kit

Manufactured for Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Carbonic Anhydrase 3 (CA3) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for CA3, with captured analyte detected using a biotinylated anti-CA3 antibody followed by an HRP-avidin conjugate. Signal is visualised with TMB substrate, producing a yellow product whose absorbance is read at 450 nm in a colorimetric microplate reader.

The assay reports a measuring range of 3.12–200 ng/ml and a sensitivity of 1.34 ng/ml, with a total incubation time of 4 h 30 min. Supplied items include a pre-coated microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, with components stored at +4°C or –20°C as instructed.

Spike-and-recovery testing (n=5) returned averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Across serial dilutions of 1:2, 1:4, 1:8 and 1:16, recoveries were respectively 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
