Human Carbohydrate Antigen 19-9 ELISA Kit

Designed for the in vitro quantitative determination of human Carbohydrate Antigen 19-9, this Antibodies.com sandwich ELISA (Enzyme-Linked Immunosorbent Assay) kit is suited for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and follows a sandwich enzyme immunoassay workflow: standards and samples are added, followed by a Biotinylated Anti-Carbohydrate Antigen 19-9 Antibody and then HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450nm in a microplate reader and the Carbohydrate Antigen 19-9 concentration is calculated by reference to a standard curve.

Performance characteristics include a detection range of 3.13-200 U/ml and a sensitivity of <1.18 U/ml. The assay is read on a colorimetric microplate reader platform and has a total assay time of 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Spike-and-recovery performance (n=5) has been reported as: serum 82-102% (average 92%), EDTA plasma 90-102% (average 96%), and heparin plasma 88-100% (average 94%). Dilution linearity has been assessed at 1:2, 1:4, 1:8, and 1:16 dilutions in serum, EDTA plasma, and heparin plasma.
