Human Carbamoyl Phosphate Synthase 1, Mitochondrial ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human CPS1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate (12 x 8 well strips), where an antibody specific for CPS1 captures the analyte from standards and test samples.

Following incubation and washing, a Biotinylated Anti-CPS1 Antibody binds the captured CPS1, and an HRP-Avidin conjugate is then added. TMB substrate produces a blue-coloured product that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of CPS1 captured, and concentration is calculated from the O.D. absorbance read at 450 nm in a colorimetric microplate reader.

The kit has a stated product range of 0.156-10 ng/ml and a sensitivity of <0.056 ng/ml, with a total assay time of 4 hours 30 minutes. Spike-and-recovery validation (n=5 per matrix) gave ranges of 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma, with dilution linearity demonstrated at 1:2, 1:4, 1:8 and 1:16.
