Antibodies.com supplies a sandwich ELISA kit for the quantitative determination of human Calumenin in tissue homogenates, cell lysates or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for Calumenin, with detection carried out using a Biotinylated Anti-Calumenin Antibody and an HRP-Avidin conjugate. Colour development is achieved with TMB substrate: the HRP enzymatic reaction yields a blue product that changes to yellow following addition of an acidic stop solution, and the O.D. absorbance is read at 450 nm in a colorimetric microplate reader. The absorbance is proportional to Calumenin concentration and is interpreted against a standard curve.

The reported assay range is 0.625-40 ng/ml, with a sensitivity of <0.215 ng/ml, and a total assay time of 4h 30m. Sample recovery has been evaluated in serum (range 80-102%, average 91%), EDTA plasma (range 81-100%, average 90%) and heparin plasma (range 80-89%, average 84%), with dilution-linearity data reported across 1:2, 1:4, 1:8 and 1:16 dilutions in each sample matrix. The plate is provided as a pre-coated 96-well microplate arranged as 12 x 8 well strips.

Supplied components include lyophilized standard, Detection Solution A and Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Storage instructions indicate that kit components should be kept at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The manufacturer notes the kit is suitable for cell lysates and tissue homogenates, and the assay is classified as a quantitative sandwich ELISA with colourmetric detection.
