Human Calpain 2, Large Subunit ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Calpain 2, Large Subunit. The kit is intended for use with tissue homogenates and other biological fluids, and is suitable for serum, EDTA plasma and heparin plasma samples. Detection is carried out on a pre-coated microplate using a biotinylated anti-Calpain 2 detection antibody and an HRP-avidin conjugate. Bound conjugate is visualised with a TMB substrate that produces a blue colour changing to yellow upon addition of an acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the Calpain 2 concentration is calculated from a standard curve.

According to the manufacturer's specification, the assay has a sensitivity of <0.054 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of 4 h 30 min. Reported mean recovery values are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 is reported as 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components are stored at +4 °C or −20 °C as instructed and the kit should not be used past its expiration date.
