Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human CAPS1 in tissue homogenates or other biological fluids. The product is supplied for research use only and is not intended for diagnostic or therapeutic applications.

Reported performance characteristics include a sensitivity of less than 0.052 ng/ml and an assay range of 0.156-10 ng/ml. Recovery data are reported for serum (80%-102%, mean 91%; n=5), EDTA plasma (81%-100%, mean 90%; n=5) and heparin plasma (80%-89%, mean 84%; n=5). Dilution linearity ranges are reported at 1:2, 1:4, 1:8 and 1:16 dilutions for serum (n=5) at 87-91%, 87-107%, 74-101% and 92-97%; for EDTA plasma (n=5) at 90-105%, 84-101%, 90-101% and 79-108%; and for heparin plasma (n=5) at 84-95%, 92-105%, 82-105% and 89-91%. The total assay time is approximately 4 hours 30 minutes, with absorbance measured at 450 nm on a colorimetric microplate reader.

The Human CAPS1 ELISA Kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for CAPS1 is immobilised on the plate; standards and samples are added and any CAPS1 present is captured. After washing, a biotinylated anti-CAPS1 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution; the intensity of the yellow colour is proportional to the amount of CAPS1 captured, and the concentration is calculated by reference to a standard curve. Kit components include a lyophilised standard (2 vials), Detection Solution A and Detection Solution B (120 µl each), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers, with reagents stored at +4°C or -20°C as instructed.
