Human Calcium/Calmodulin Dependent Protein Kinase II Alpha ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of human Calcium/Calmodulin Dependent Protein Kinase II Alpha (CaMKII alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids [S1]. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells where CaMKII alpha binds the immobilised capture antibody; a biotinylated anti-CaMKII alpha antibody then binds the captured target, and an HRP-Avidin conjugate with TMB substrate produces a blue product that turns yellow after the acidic stop solution. Absorbance is read at 450 nm and the concentration is calculated from the standard curve [S1].

The standard curve product range is 0.156-10 ng/ml and the kit sensitivity is <0.064 ng/ml [S1]. Total assay time is 4h 30m [S1]. Spike-and-recovery experiments gave 83%-100% (average 91%) in serum (n=5), 90%-98% (average 94%) in EDTA plasma (n=5) and 90%-99% (average 94%) in heparin plasma (n=5) [S1]. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 90-99%, 82-108%, 82-96% and 92-103% for serum; 84-105%, 88-94%, 89-107% and 72-106% for EDTA plasma; and 89-92%, 88-108%, 84-102% and 90-98% for heparin plasma, all measured at n=5 [S1]. The assay is read on a colorimetric microplate reader, with kit components stored at +4°C or -20°C as instructed in the protocol and not used past the expiration date [S1].
