Human Cadherin, Neuronal ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human N Cadherin in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for N Cadherin immobilised on the plate. Standards and test samples are added, followed by a Biotinylated Anti-N Cadherin Antibody and an HRP-Avidin conjugate. A TMB substrate produces a blue colour that turns yellow after addition of an acidic stop solution; absorbance is read at 450 nm in a microplate reader and the N Cadherin concentration is calculated from the standard curve.

According to the manufacturer's documentation, the assay range is 0.156–10 ng/ml and the sensitivity is <0.054 ng/ml. Reported recoveries with n=5 are: serum 80–102% (average 91%), EDTA plasma 81–100% (average 90%), and heparin plasma 80–89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions is reported as 87–91%, 87–107%, 74–101%, 92–97% for serum; 90–105%, 84–101%, 90–101%, 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, 89–91% for heparin plasma. The kit includes a pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components are stored at +4°C or −20°C as instructed, and total assay time is 4 hours 30 minutes.
