Human Cadherin, Heart ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human H Cadherin in serum, plasma, tissue homogenates, and other biological fluids. A microtiter plate has been pre-coated with an antibody specific for H Cadherin. Standards and samples are added into the wells and H Cadherin present in each sample is bound to the wells by the immobilized antibody. Following incubation, the wells are washed and then incubated with a biotinylated anti-H Cadherin antibody, which binds the captured H Cadherin. HRP-Avidin conjugate is then added to the wells and the microtiter plate is incubated. TMB substrate solution is then used to visualize the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of H Cadherin captured in each well. The concentration of H Cadherin is calculated by reading the absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a sensitivity of 0.058 ng/ml and a range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Detection is colourimetric. Validation data are reported for serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%) and heparin plasma (n=5, range 80%-89%, average 84%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Components are stored at +4°C or -20°C as instructed.
