The assay uses a pre-coated microtiter plate with an immobilised antibody specific for C1QTNF9; standards and samples are added, followed by a biotinylated anti-C1QTNF9 detection antibody, an HRP-avidin conjugate and a TMB substrate. The enzymatic reaction produces a blue product that turns yellow after addition of acidic stop solution, and the absorbance is read at 450 nm in a colorimetric microplate reader with concentration calculated against a standard curve.

According to the manufacturer, the assay sensitivity is <1.32 ng/ml and the working range is 3.12–200 ng/ml, with a total protocol time of approximately 4 hours 30 minutes.
