From Antibodies.com, this competitive enzyme-linked immunosorbent assay (cELISA) provides the in vitro quantitative determination of human C-Peptide in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay employs the competitive enzyme immunoassay technique, in which C-Peptide present in samples or standards competes with a fixed amount of immobilised C-Peptide on the pre-coated microtiter plate for binding sites on a biotinylated anti-C-Peptide antibody. Unbound antibody is removed by washing before an HRP-avidin conjugate is added.

A TMB substrate solution visualises the HRP enzymatic reaction as a blue product that changes to yellow after addition of acidic stop solution. The density of yellow is inversely proportional to the amount of C-Peptide in each sample or standard, and the concentration is read at 450 nm in a microplate reader against a standard curve. The stated working range is 0.312–20 ng/ml with a sensitivity of <0.148 ng/ml, and the full assay is completed in approximately 2h 30m. Detection is colourimetric on a pre-coated microplate platform.

Reported average recoveries are 91% for serum, 90% for EDTA plasma, and 93% for heparin plasma, with supporting dilution linearity data for each matrix. Kit components are stored at +4 °C or −20 °C as instructed and the kit should not be used past its expiration date.
