Description

Manufactured by Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) is intended for the in vitro quantitative determination of human Bridging Integrator 2 (BIN2) in tissue homogenates, cell lysates, and other biological fluids. The assay is read on a colorimetric microplate reader.

The test uses a 96-well microplate pre-coated with an antibody specific for BIN2. Standards and test samples are added to the wells, where BIN2 is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-BIN2 Antibody binds the captured BIN2, followed by addition of HRP-Avidin conjugate. TMB substrate solution is then used to visualize the reaction, producing a blue-coloured product that turns yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of BIN2 captured in each well, and concentration is calculated by reading the absorbance at 450 nm against the standard curve. Total assay time is 4h 30m.

Reported sensitivity is less than 0.112 ng/ml with an assay range of 0.312–20 ng/ml. Validation data report the following recovery ranges in spiked samples: serum (n=5) 80–102% with an average of 91%; EDTA plasma (n=5) 81–100% with an average of 90%; and heparin plasma (n=5) 80–89% with an average of 84%. Dilution linearity (at 1:2, 1:4, 1:8 and 1:16) is reported as 87–91%, 87–107%, 74–101%, 92–97% for serum (n=5); 90–105%, 84–101%, 90–101%, 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105%, 89–91% for heparin plasma (n=5). Kit components should be stored at +4°C or –20°C as instructed and not used past the expiration date.
