Human Breast Cancer Susceptibility Protein 2 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human BRCA2 in tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated 96-well microplate on which an antibody specific for BRCA2 immobilises the target. Standards and test samples are added to the wells, and bound BRCA2 is detected using a Biotinylated Anti-BRCA2 Antibody followed by an HRP-Avidin conjugate and TMB substrate. The resulting colour change is read at 450 nm on a colorimetric microplate reader and the concentration of BRCA2 is calculated by reference to the standard curve.

The stated detection range covers 0.312-20 ng/ml with a sensitivity of <0.107 ng/ml, and the complete assay time is 4h 30m. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Average recovery in serum (n=5) was 91% with a range of 80-102%, in EDTA plasma (n=5) 90% with a range of 81-100%, and in heparin plasma (n=5) 84% with a range of 80-89%. Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 was 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
