Human Breast Cancer Susceptibility Protein 1 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Breast Cancer Susceptibility Protein 1 (BRCA1) in tissue homogenates, cell lysates and other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, a biotinylated anti-BRCA1 detection antibody and an HRP-Avidin conjugate, with signal generated by a TMB substrate solution. The enzymatic reaction is read at 450 nm on a colorimetric microplate reader and BRCA1 concentrations are calculated by reference to a standard curve.

The total assay time is 4 hours 30 minutes, with a reported sensitivity of <15.6 pg/ml and a detection range of 31.2–2000 pg/ml. Standards and samples are added to the antibody-coated wells where BRCA1 is captured; following incubation and washing, the biotinylated detection antibody binds the immobilised analyte. After addition of HRP-Avidin conjugate and further washing, TMB substrate produces a blue-coloured product that changes to yellow upon addition of the acidic stop solution, with the intensity of yellow being directly proportional to the amount of BRCA1 captured in each well.
