From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables the in vitro quantitative determination of human BCAR1 in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips), with an antibody specific for BCAR1 immobilised on the wells. Standards and samples are added, followed by a Biotinylated Anti-BCAR1 Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised using TMB substrate solution, and absorbance is read at 450 nm on a colorimetric microplate reader, with BCAR1 concentration calculated against the standard curve.

Sensitivity is <0.053 ng/ml and the assay range is 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Kit components are stored at +4°C or –20°C as instructed. Recovery values (n=5) are 91% for serum (range 80–102%), 90% for EDTA plasma (81–100%) and 84% for heparin plasma (80–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 (n=5) was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
