Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Bone Morphogenetic Protein 7 (BMP7) in serum, plasma, tissue homogenates, and other biological fluids. The procedure uses a pre-coated 96-well microplate configured as 12 x 8 well strips, with an antibody specific for BMP7 immobilised on the plate to capture analyte from standards and samples. A biotinylated anti-BMP7 detection antibody is added, followed by HRP-Avidin conjugate, and TMB substrate produces a blue product that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450 nm on a colorimetric microplate reader and the BMP7 concentration is calculated against the standard curve.

Antibodies.com reports a product range of 62.5–4000 pg/ml with a sensitivity of 23.4 pg/ml. The assay is a sandwich (quantitative) ELISA with a total run time of 4 hours 30 minutes. Validated recovery is 80%–102% (average 91%) in serum (n=5), 81%–100% (average 90%) in EDTA plasma (n=5), and 80%–89% (average 84%) in heparin plasma (n=5). Dilution linearity data are reported at 1:2, 1:4, 1:8, and 1:16 dilutions for all three sample types.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solution A, detection solution B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components are stored at +4°C or -20°C as instructed in the product manual, and the kit should not be used past its expiration date.
