This sandwich ELISA kit from Antibodies.com is intended for the in vitro quantitative determination of human BST2/Tetherin (Bone Marrow Stromal Cell Antigen 2) in tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for BST2/Tetherin that has been pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where the target is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-BST2/Tetherin Antibody is added and binds the captured target, followed by incubation with an HRP-Avidin conjugate. Following further incubation and washing, TMB substrate solution is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow colour is proportional to the amount of BST2/Tetherin captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a quantitative detection range of 0.156-10 ng/ml and a sensitivity of <0.068 ng/ml. Total assay time is 4 hours 30 minutes, with detection performed on a colorimetric microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Validation data demonstrate the following recovery ranges and averages (n=5): serum 80-102% (average 91%), EDTA plasma 81-100% (average 90%), and heparin plasma 80-89% (average 84%). Linearity upon dilution has been assessed at 1:2, 1:4, 1:8 and 1:16, with corresponding recovery ranges for serum of 87-91%, 87-107%, 74-101% and 92-97%; for EDTA plasma of 90-105%, 84-101%, 90-101% and 79-108%; and for heparin plasma of 84-95%, 92-105%, 82-105% and 89-91%.
