Human Beta-Carotene-15,15'-Monooxygenase 1 ELISA Kit

Antibodies.com supplies this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) for the in vitro quantitative determination of human Beta-Carotene-15,15'-Monooxygenase 1 (BCO1) in tissue homogenates, cell lysates and other biological fluids. The kit is provided on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with results read colourimetrically at 450 nm in a standard microplate reader.

An antibody specific for BCO1 is immobilised on the plate; standards and test samples are added and bound BCO1 is detected using a Biotinylated Anti-BCO1 Antibody followed by an HRP-Avidin conjugate. The TMB substrate yields a yellow product upon addition of the acidic stop solution, the absorbance of which is proportional to BCO1 concentration and is quantified against a standard curve. Reported sensitivity is <0.103 ng/ml with an assay range of 0.312–20 ng/ml, and the total assay time is approximately 4 hours 30 minutes. Components are stored at +4 °C or –20 °C as instructed.

Recovery and linearity have been characterised for serum, EDTA plasma and heparin plasma. Average recoveries (n=5) were 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%), with linearity values reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each matrix.
