Human Beclin 1 ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human Beclin 1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for Beclin 1. Standards and test samples are added to the wells, where Beclin 1 is captured by the immobilized antibody, followed by incubation with a Biotinylated Anti-Beclin 1 Antibody and an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of Beclin 1 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

The assay has a standard range of 0.156-10 ng/ml and a sensitivity of 0.059 ng/ml. It is a quantitative colorimetric sandwich ELISA with a total assay time of 4 hours 30 minutes run on a pre-coated microplate platform of 12 x 8 well strips. Suitable sample types include serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Average recovery values reported are 91% for serum (range 80%-102%, n=5), 90% for EDTA plasma (range 81%-100%, n=5), and 84% for heparin plasma (range 80%-89%, n=5). Linearity across 1:2, 1:4, 1:8 and 1:16 dilutions falls within 74-108% across the tested sample types.
