An antibody specific for the target is pre-coated onto a microtiter plate; standards and samples are added, followed by a Biotinylated Anti-MIST-1 Antibody and an HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow upon addition of stop solution, with absorbance read at 450 nm in a microplate reader.

The kit reports a sensitivity of 0.071 ng/ml and an assay range of 0.156-10 ng/ml. Total assay time is 4 hours 30 minutes, with colourmetric detection on a pre-coated microplate platform.
