Human Basic Fibroblast Growth Factor ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com and intended for the in vitro quantitative determination of human Basic Fibroblast Growth Factor (bFGF) in serum, plasma, urine, cell culture supernatant, and other biological fluids. The assay applies a quantitative sandwich immunoassay in which standards or samples are incubated in microplate wells pre-coated with a monoclonal antibody specific for bFGF, followed by addition of a standardised horseradish peroxidase (HRP)-conjugated antibody to "sandwich" the immobilised analyte. After washing, a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added, the enzyme–substrate reaction is terminated with a sulphuric acid solution, and the colour change is measured spectrophotometrically at 450 nm ± 2 nm. Two calibrator diluents are included: Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant/urine testing, allowing the operator to produce a standard curve of optical density versus bFGF concentration in pg/mL.

The assay exhibits no detectable cross-reactivity with human TGF-β, GM-CSF, MCP-3, INF-r, EGF, IL-1a, IL-8, IL-1ß, TNF-a, IL-16, MCSF, IL-5, and IL-16, and recognises both natural and recombinant human bFGF. Reported sensitivity is 78 pg/ml, with a product range of 78–8000 pg/ml. Detection is colorimetric on a pre-coated microplate platform configured as 12 x 8 well strips. The kit is shipped and stored at 4°C and is intended to be read on a colorimetric microplate reader.
