Description

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Livin (Baculoviral IAP Repeat Containing Protein 7) in serum, plasma, tissue homogenates, cell lysates and other biological fluids. The assay is read on a colorimetric microplate reader and runs for 4h 30m using a pre-coated 96-well microplate arranged as 12 x 8 well strips. Sensitivity is reported as less than 0.073 ng/ml, with an assay range of 0.156-10 ng/ml.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for Livin is pre-coated onto the microplate, capturing Livin from standards and test samples. After washing, a Biotinylated Anti-Livin Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then added, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of Livin captured in each well, with concentration calculated by reading O.D. absorbance at 450 nm and referring to the standard curve.

Recovery has been characterised in serum (n=5) at 80%-102% (average 91%), EDTA plasma (n=5) at 81%-100% (average 90%) and heparin plasma (n=5) at 80%-89% (average 84%), with dilution linearity examined at 1:2, 1:4, 1:8 and 1:16 in each matrix. Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
