Human B-Lymphoid Tyrosine Kinase ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) enables the in vitro quantitative determination of human B Lymphoid Tyrosine Kinase (BLK) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated microplate to capture BLK from standards and samples, followed by incubation with a Biotinylated Anti-BLK Antibody and then an HRP-Avidin conjugate. TMB substrate is added to visualise the enzymatic reaction, which is stopped with an acidic stop solution; absorbance is read at 450nm on a colorimetric microplate reader and BLK concentration is calculated from the standard curve.

The kit reports an assay range of 0.156–10 ng/ml, a sensitivity of <0.071 ng/ml and a total assay time of 4h 30m. Validation across serum, EDTA plasma and heparin plasma samples (n=5 each) showed mean recoveries of 91%, 90% and 84% respectively. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16, with individual recoveries spanning 74–108% across the matrices. Kit components are stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.
