From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Ctip1/BCL-11A (B Cell CLL/Lymphoma 11A) in tissue homogenates, cell lysates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for Ctip1/BCL-11A pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where Ctip1/BCL-11A is captured by the immobilised antibody. After washing, a Biotinylated Anti-Ctip1/BCL-11A Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450nm in a microplate reader and the concentration of Ctip1/BCL-11A is calculated by reference to a standard curve.

The kit has a reported sensitivity of <0.109 ng/ml with an assay range of 0.312–20 ng/ml, and a total assay time of approximately 4 hours 30 minutes. Detection is colourimetric and the assay is intended for use with a colorimetric microplate reader. Recovery has been characterised in serum (83–100%, n=5, average 91%), EDTA plasma (90–98%, n=5, average 94%) and heparin plasma (89–102%, n=5, average 95%). Linearity has also been demonstrated across 1:2 to 1:16 dilutions in the same matrices (serum 79–110%, EDTA plasma 73–109%, heparin plasma 77–110%). Kit components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
