Human B-Cell Activation Factor Receptor ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of human BAFF-R in tissue homogenates, cell lysates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for BAFF-R pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells and BAFF-R is captured by the immobilised antibody, followed by incubation with a Biotinylated Anti-BAFF-R Antibody and HRP-Avidin conjugate. TMB substrate is added to visualise the reaction; the resulting yellow colour intensity is proportional to BAFF-R concentration and is read at 450 nm on a colorimetric microplate reader against a standard curve.

The assay has a sensitivity of <0.061 ng/ml and a working range of 0.156-10 ng/ml, with a total assay time of 4h 30m. The platform is a pre-coated 96-well microplate arranged as 12 x 8 well strips. Supplied components include a Pre-Coated 96 Well Microplate, Lyophilized Standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers. Storage is at +4°C or -20°C as instructed for individual components; the kit should not be used past its expiration date.

Recovery has been characterised in serum (n=5, 80-102%, average 91%), EDTA plasma (n=5, 81-100%, average 90%) and heparin plasma (n=5, 80-89%, average 84%). Parallel performance data are reported at 1:2, 1:4, 1:8 and 1:16 dilutions across the same sample types, with serum ranging 87-91%, 87-107%, 74-101% and 92-97%, EDTA plasma 90-105%, 84-101%, 90-101% and 79-108%, and heparin plasma 84-95%, 92-105%, 82-105% and 89-91%.
