Human B-Cell Activating Factor ELISA Kit

A sandwich-format immunoassay kit from Antibodies.com designed for the in vitro quantitative determination of human BAFF in serum, plasma, tissue homogenates and other biological fluids. The assay employs a capture antibody specific for BAFF pre-coated onto a microtiter plate; standards and samples are added, followed by a biotinylated anti-BAFF detection antibody and an HRP-avidin conjugate. The enzymatic reaction is visualised using TMB substrate, producing a colour change that is read at 450 nm in a microplate reader, with BAFF concentration calculated against the standard curve. Sensitivity is reported as <0.055 ng/ml and the quantitative range is 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes.

Spike-and-recovery has been validated in serum (range 80–102%, average 91%), EDTA plasma (range 81–100%, average 90%) and heparin plasma (range 80–89%, average 84%). Linearity of dilution has also been examined across 1:2 to 1:16 sample dilutions in the same matrices, with reported recovery values generally falling between 74% and 108%.
