Manufactured for Antibodies.com, this sandwich enzyme-linked immunosorbent assay (ELISA) is designed for the in vitro quantitative determination of human Autoimmune Regulator (AIRE) in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for AIRE is pre-coated onto a microtiter plate. Standards and samples are added, followed by washing and incubation with Biotinylated Anti-AIRE Antibody and then HRP-Avidin conjugate. A TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader, with AIRE concentration calculated against a standard curve.

The kit reports a sensitivity of <0.053 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Manufacturer-supplied average recovery values are 91% (range 80%-102%, n=5) in serum, 90% (range 81%-100%, n=5) in EDTA plasma, and 84% (range 80%-89%, n=5) in heparin plasma. Linearity across dilutions of 1:2, 1:4, 1:8, and 1:16 shows serum recovery ranges of 87-91%, 87-107%, 74-101%, and 92-97% respectively; EDTA plasma ranges of 90-105%, 84-101%, 90-101%, and 79-108%; and heparin plasma ranges of 84-95%, 92-105%, 82-105%, and 89-91%. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its stated expiration date.
