Description
A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human ATP Binding Cassette Transporter G1 (ABCG1) in tissue homogenates, cell lysates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate, with an antibody specific for ABCG1 immobilised on the plate. Standards and samples are added to the wells; captured ABCG1 is bound by a biotinylated anti-ABCG1 antibody, and detection is carried out using an HRP-avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that turns yellow after addition of an acidic stop solution, with absorbance measured at 450 nm on a colorimetric microplate reader. ABCG1 concentration in each sample is calculated from a standard curve.
Quoted performance figures from the manufacturer datasheet include a sensitivity of 0.061 ng/ml and an assay range of 0.156–10 ng/ml. The total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed and must not be used past the stated expiration date. Reported recovery in spiked samples averages 91% in serum (range 80%–102%, n=5), 90% in EDTA plasma (range 81%–100%, n=5) and 84% in heparin plasma (range 80%–89%, n=5).
