Human ATP Binding Cassette Transporter C2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of human ATP Binding Cassette Transporter C2 (MRP2) in tissue homogenates, cell lysates or other biological fluids. The method uses the sandwich enzyme immunoassay technique, with an antibody specific for MRP2 pre-coated onto a 96-well microtiter plate. Standards and test samples are added and the MRP2 present binds to the immobilised antibody; after incubation and washing, a Biotinylated Anti-MRP2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution visualises the HRP enzymatic reaction as a blue colour turning yellow on addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of MRP2 captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against the standard curve.

The kit is reported with a sensitivity of <0.067 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Validation data referenced for the product lists serum recovery at an average of 91% (n=5, range 80–102%), EDTA plasma at 90% (n=5, 81–100%) and heparin plasma at 84% (n=5, 80–89%). Detection is colourimetric on a pre-coated microplate platform, and the kit is described as suitable for cell lysates and tissue homogenates. Kit components are stored as instructed, with some components stored at +4°C and others at -20°C; the kit should not be used past its expiration date.
