The Antibodies.com Human Aspartate Aminotransferase ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human Aspartate Aminotransferase in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Aspartate Aminotransferase pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, followed by a Biotinylated Anti-Aspartate Aminotransferase Antibody and an HRP-Avidin conjugate. A TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of stop solution. The density of yellow is proportional to the amount of Aspartate Aminotransferase captured, and concentration is calculated from the O.D. absorbance at 450 nm using a colorimetric microplate reader.

According to the manufacturer, the assay has a product range of 0.312–20 ng/ml and a sensitivity of <0.123 ng/ml. Total assay time is 4 hours 30 minutes. Spike-and-recovery results were reported for serum (range 80%–102%, average 91%, n=5), EDTA plasma (81%–100%, average 90%, n=5) and heparin plasma (80%–89%, average 84%, n=5). Linearity upon dilution (1:2, 1:4, 1:8 and 1:16) was reported across serum, EDTA plasma and heparin plasma samples.

Kit components include the pre-coated 96-well microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage at +4°C or -20°C as instructed. The components must not be used past the expiration date.
